I am swapping a 5.7 vortec L31 with an 0411 into an s10. The 5.7 one wire knock sensor will not clear the motor mount but I can mount the 4.3 vortec 2 wire knock sensor, is there a way to use the 2 wire knock sensor with the 02 express van tune?

Yes ... but you need a different tuning software to configure the tune. In an S10 swap some have mounted the knock sensor to the block side motor mount (plate steel) - as a work around.
