Trans relearn after tune file upload

Is there any validity to doing a relearn to reset CVI after uploading a new tune file for transmission specific tuning? I just went through a few changes without doing a relearn and now I'm back at square one with the stock tune file. I did not like how hard the shifts were. This is a 66RFE behind a 5.7 hemi. I'm also in the learning stages here so if anyone has any pointers it would help a lot. I'm not opposed to recommendations and paying a reputable tuner who understands 66RFE, this transmission has never been right and I need it to work. I currently have a built unit in the truck with updated billet valve body, a high pressure tuneless pump and billet center support.
